DNAX-activating protein 12 (DAP12, TYROBP) is a signaling adaptor for several pathogen receptors expressed by cells of the innate immune system. The DAP12 protein structure consists of a short extracellular domain, a transmembrane domain, and a cytoplasmic immunoreceptor tyrosine-based activation motif (ITAM). DAP12 protein is expressed by hematopoietic cells, including NK cells, monocytes, macrophages, dendritic cells, mast cells, basophils, eosinophils, neutrophils, and some γδ T cells and NKT cells. DAP12 exists as a homodimer that associates with a variety of receptors involved in pathogen detection, including the KIR family of NK cell receptors. Ligand binding by DAP12-associated receptors results in phosphorylation of tyrosine residues within the DAP12 ITAM by Src family kinases and leads to activation of Syk or Zap-70 and downstream signaling responses. Describe :

Blocking peptides are peptides that bind specifically to the target antibody and block antibody binding. These peptide usually contains the epitope recognized by the antibody. Antibodies bound to the blocking peptide no longer bind to the epitope on the target protein. This mechanism is useful when non-specific binding is an issue, for example, in Western blotting (WB) and Immunohistochemistry (IHC). By comparing the staining from the blocked antibody versus the antibody alone, one can see which staining is specific; Specific binding will be absent from the western blot or IHC performed with the neutralized antibody.
