Rho, the Ras-related small GTPase, is responsible for the regulation of Actin-based cytoskeletal structures, including stress fibers, focal adhesions and the contractile ring apparatus. Rho proteins function as molecular switches that are able to turn cytokinesis on and off. Although little is known about signaling downstream of Rho, a host of putative Rho effector proteins have been de-scribed, including rhophilin, Rhotekin, citron and the serine/threonine kinase, protein kinase N. Two additional Rho-activated serine/threonine kinases have been described, designated Rock-1 and Rock-2 (also referred to as Roka) for Rho-associated coil-containing protein kinase. Western blot analysis of ROCK2 on Raji (1) and SiHa (2) cell lysates using anti-ROCK2 antibody at 1/500 dilution. -
ICC staining ROCK2 in Hela cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Blocking peptides are peptides that bind specifically to the target antibody and block antibody binding. These peptide usually contains the epitope recognized by the antibody. Antibodies bound to the blocking peptide no longer bind to the epitope on the target protein. This mechanism is useful when non-specific binding is an issue, for example, in Western blotting (WB) and Immunohistochemistry (IHC). By comparing the staining from the blocked antibody versus the antibody alone, one can see which staining is specific; Specific binding will be absent from the western blot or IHC performed with the neutralized antibody.Formula:
